Assay principle: sandwich

AQP4 molecules are coated onto the ELISA plate wells. AQP4-biotin complexes constitute the liquid phase. AQP4 Ab from the sample will bind to the AQP4 coated on the well and interact with AQP4-biotin to form a bridge. The bound AQP4-biotin is detected through the addition of streptavidin peroxidase (SA-POD). The unbound SA-POD in excess is washed off before addition of the peroxidase substrate TMB.

Assay procedure: 2h at RT / shaking + 20 min at RT / shaking + 20 min at RT in the dark.

Sample: serum – 50 µL

Analytical sensitivity: 0.17 U/mL

Normal values

Samples from 216 presumably healthy blood donors were analyzed. 100% were identified as being negative for AQP4 autoantibodies.
