Assay principle: sandwich

GAD65 and IA-2 molecules are coated onto the ELISA plate wells. GAD65-biotin and IA-2-biotin complexes constitute the liquid phase. GAD-Ab and IA-2-Ab from the sample bind to the GAD65 and to IA-2 immobilized onto the well and interact with GAD65-biotin and IA-2 biotin via a GAD-Ab and IA-2-Ab bridges. Addition of streptavidin peroxidase detects the bound GAD65-biotin and IA-2 biotin.

The unbound streptavidin peroxidase in excess is washed off before the addition of the peroxidase substrate TMB. Assay procedure: 16-20h at 2-8°C + 1h at RT / shaking + 20 min at RT / shaking + 20 min at RT in the dark.
